Funny I just got done shooting the GFX-50S with an old Mamiya 645 manual focus lens with aperture ring a few minutes ago - a very similar configuration to what you are asking about. With the adapter and lens and the camera set to MF mode with peaking on, it works very well.

The 100mm macro for the 6x7 is wickedly sharp, but manual focus only. And not very inexpensive either. The latest 55mm 67 lens is also extremely sharp, but a tad wide for some people' tastes. I'd read reviews of the autofocus version of my 45-85mm which have said that it is sharper than the primes for the Pentax 645n.

Manual focus requires more accuracy than AF from the user. To further compound the problem (but still related to accuracy) is the larger format itself: the DOF is less with MF and LF. When focusing the 600/5.6 lens on the 645D at 5m (it' closest focusing distance), the DOF is less than 1/4 inch.
